Research in this lab lies at the interface of chemistry and biology and spans a broad range of topics including mechanistic enzymology, cofactor biogenesis, radical enzymology, and metalloprotein-mediated redox stress-linked transcriptional activation and gene regulation mechanisms. The long-term goal of our studies is to determine the chemical basis for the biological roles and physiological effects of metal ions and protein-based free radicals. Broadly, the two primary themes of our research have been (1) the investigation and characterization of transient intermediates of metalloprotein-mediated reactions and (2) the modulation of signal transduction by metalloproteins. In order to pursue these goals, methods from molecular biology to physical chemistry are utilized: cloning, site-directed mutagenesis, column chromatography, steady-state and rapid kinetics, protein X-ray crystallography, and electron paramagnetic resonance spectroscopy. Our work furthers the understanding of enzyme mediated reactions through the elucidation of reaction mechanisms and protein structures and helps to understand structure-function relationships in enzymes and metalloproteins.
